Conjugated antibodies are antibodies that have been joined or conjugated to a type of second molecule. The second molecule can be a toxin, radioisotope, or other label. Abcore-Inc. offers numerous conjugated antibodies come labeled with a fluorescent dye and proteins. Abcore-Inc provides antibodies conjugated to Alexa Fluor® 700, Alexa Fluor® 647, Alexa Fluor® 488, APC, FITC, Pacific Blue™ , Pacific Orange™, Dyomics® 547, and Dyomics® 647, PE, biotin, PerCP, and Peroxidase. Conjugated antibodies are also called immonoconjugates and are mostly used in immunotherapy or in development of antibodies used to target specific tissues. Fluorescent dyes can be tagged to antibodies and used in applications like flow cytometry, fluorescent microscopy, immunohistochemistry, primary detection, ELISAs and immunocytochemistry.
Alexa Fluor® offers a family of fluorescent dyes for immunoconjugates. APC stands for allophycocyanin. It is another fluorescent probe for conjugation. FITC is a common abbreviation for fluorescein. It is a small organic molecule that can be attached to antibodies. Pacific Blue™ and Pacific Orange™ antibodies are tagged with excitable dyes. Dyomics®547 and Dyomics®647 are succinimide esters that can directly be conjugated to antibodies and allow them to fluoresce in a variety of colors. Phycoerythrin or PE is a commonly used fluorochrome of rather large size isolated for algae. Biotin conjugation is useful because it binds to streptavidin and avidin with an extremely high affinity and these molecules are resistant to high heat, pH, and proteolysis. Peridinin Chlorophyll Protein Complex is also known as PerCP. PerCP is a water soluble carotenoid pigment found in photosynthetic dinoflagellates. PerCP conjugated antibodies are typically used in flow cytometry experiments. Horseradish peroxidase (HRP) is an enzyme found in horseradish that is conjugated to antibodies in order to amplify a weak signal and increase the chance of the target molecule being detected.
Anti-NTAL antibody (Alexa Fluor®488) binds against the NTAL target. NTAL is expressed in the cell membrane and lipid rafts in cells of the spleen, peripheral blood lymphocytes, germinal centers of lymph nodes, placenta, lung, pancreas, small intestine, and B-cells. NTAL protein is involved with the high affinity immunoglobulin epsilon receptor FCER1. It is thought to be involved in B-cell antigen receptor mediated signaling in B-cells and FCGR1 mediated signaling in myeloid cells. Defects or deletions in the gene encoding for NTAL can be the cause of Williams-Beuren syndrome (WBS). William’s syndrome is a rare genetic condition that is characterized by cardiovascular disease, developmental delays, learning disabilities, unique facial features, and highly social personalities. Anti-albumin antibody (biotin) is conjugated to biotin and binds against serum albumin. Albumin is secreted by the body and is the main protein of blood plasma. Albumin is produced in the liver. Albumin is a transport protein that carries hormones and fatty acids. Mutations in the gene that encodes for the body’s ability to produce albumin can result in different anomalous proteins which may or may not have appropriate function.
